Why do Landrace pig lymphocytes appear more susceptible to PCV2 than Piétrain lymphocytes?

Landrace pig T-lymphoblasts were more permissive to PCV2 than Piétrain cells, with endolysosomal trafficking differences that may contribute to cellular susceptibility.

Landrace pig T-lymphoblasts were more permissive to PCV2 than Piétrain cells, with endolysosomal trafficking differences that may contribute to cellular susceptibility.

Problem: Breed-associated differences in PCV2 susceptibility have been observed, but the cellular mechanisms remain poorly understood.

Methods: T-lymphoblasts from three Landrace and three Piétrain pigs were exposed to six PCV2 strains and examined for viral entry, endolysosomal trafficking, pH dependence, and protease activity.

1. Replication was generally more efficient in Landrace lymphoblasts and also varied among viral strains.

2. Blocking acidification reduced infection in Landrace cells, while inhibiting serine proteases markedly reduced infection in both breeds.

3. At 360 minutes, PCV2-lysosome colocalization reached 56.3% in Landrace versus 27.2% in Piétrain cells, but this cell-based study does not show that breed choice alone would prevent PCV2-associated disease on farms.
